Non-metabolic role of UCK2 links EGFR-AKT pathway activation to metastasis enhancement in hepatocellular carcinoma

Up-regulation of Uridine-cytidine kinase 2 (UCK2), a rate-limiting enzyme of the pyrimidine salvage pathway, has been suggested in HCC, but the detailed molecular mechanisms and therapic role of UCK2 remain elusive. Bioinformatic analyses revealed that UCK2 might be a key up-regulated metabolic gene in HCCs. The expressional pattern and prognostic value of UCK2 were further examined in a large number of clinical samples. Functional assays based on site-directed mutagenesis showed that UCK2 promoted cell proliferation in a metabolic manner, but non-catalytically facilitates HCC metastasis. Mechanistically, in response to EGF, UCK2 interacted with EGFR to block EGF-induced EGFR ubiquitination and degradation, which resulted in elevated EGFR-AKT pathway activation and metastasis enhancement in HCCs. Concurrent pharmacological targeting on UCK2 and EGFR showed synergistic effects on HCC treatment. This study disclosed the non-metabolic role of UCK2 and suggested the therapeutic potential of concurrent blocking the metabolic and non-metabolic roles of UCK2 in HCC treatment.

Effect of homonuclear boron bonds in the adsorption of DNA nucleobases on boron nitride nanosheets

Quantum-mechanics calculations were carried out within the density functional theory (DFT) scheme, to analyze the sorbate-sorbent interaction between DNA nucleobases (purines: guanine and adenine; pyrimidines: cytosine and thymine) and hexagonal boron nitride nanosheets (pristine and non-stoichiometric) in two phases: i) gas and ii) aqueous. The resulting molecular simulations for the pristine nanosheet indicate that the four molecular sorbates prefer to be oriented perpendicular and/or parallel respect to sorbent. This geometrical effect generates adsorption energies associated to non-covalent interactions (physisorption), being the preferential adsorption sites those of N atoms of the nanosheet. According to the calculated quantum descriptors, they exhibit low chemical reactivity and work function, as well as high polarity and semiconductor-like behavior. However, the homonuclear boron bonds in the nanosheet (negatively charged) induce a strong interaction, almost three times larger than pristine nanosheet in gas phase; except for cytosine, due to this is weakly adsorbed in both phases. Moreover, the chemical reactivity and work function are reduced, whereas its conductivity (energy LHgap) and polarity were increased, since the preferential interaction site is corresponding to a B atom. Pyrazolo[3,4-d]pyrimidines represent an important class of heterocyclic compounds well-known for their anticancer activity exerted by the inhibition of eukaryotic protein kinases. Recently, pyrazolo[3,4-d]pyrimidines have become increasingly attractive for their potential antimicrobial properties. Here, we explored the activity of a library of in-house pyrazolo[3,4-d]pyrimidines, targeting human protein kinases, against Staphylococcus aureus and Escherichia coli and their interaction with ampicillin and kanamycin, representing important classes of clinically used antibiotics. Our results represent a first step towards the potential application of dual active pyrazolo[3,4-d]pyrimidine kinase inhibitors in the prevention and treatment of bacterial infections in cancer patients.
