Streptococcus pneumoniae is a natural colonizer of the human respiratory tract and an opportunistic pathogen. Although epithelial cells are among the first to encounter pneumococci, the cellular processes and contribution of epithelial cells to the host response are poorly understood. To provide evidence on involvement of JMJD3 demethylase in prostate tumor progression, & particularly on MGMT, TRA2A, RPS6KA2, & U2AF1 candidate gene expressions, anal. of H3K27me3 enrichment was performed by chromatin immunoprecipitation (ChIP)quant. polymerase chain reaction (qPCR) after inhibition of JMJD3 by GSK-J4 in prostate cancer cell lines. Subsequently, reverse transcription (RT)-qPCR conducted after inhibition of JMJD3 by siRNA/GSK-J4 inhibitor demonstrating its demethylase activity on expression of studied genes. In addn, inhibition of JMJD3 by siRNA was validated by Western blotting. Results: gene expression by RTqPCR anal. after inhibition of JMJD3 by siRNA & chem. inhibition by GSK-J4. JMJD3 inhibition by GSK-J4 exhibited non-vital decrease in JMJD3 expression in the 3 cell lines LNCaP, DU-145 & PC-3. The mRNA anal. confirmed that enrichment in H3K27me3 on target genes, led to an inhibition of MGMT, TRA2A, RPS6KA2, & U2AF1 exprsn, affirming H3K27me3 as a repressive epigenetic marker. Conclusion: The data anal. emphasized the link between JMJD3 and regulation of these genes in prostate cancer cell lines. JMJD3, regulated in vitro by siRNA/GSK-J4, making JMJD3 a potential therapeutic target. In future, it would be interesting to see effect of JMJD3 inhibition on those genes in vivo. Forskolin is a natural cAMP elevating agent that is emerging as one of the most promising mols. for its potential use in cancer therapy. The epigenetic marks play a relevant role in several cancer types, including in leukemia. In particular, GSKJ4 has been recently demonstrated to act as a potent proliferation inhibitor in many tumor cell lines. Due to the failure of the single agent therapy, recently the chemotherapy combination has received more attention in order to increase the therapeutic index and to reduce the side effects. In this scenario, naturally occurring mols. represent the ideal candidates to investigate cooperative responses with conventional and new antineoplastic drugs. Here, we investigate the capability of forskolin to potentiate the effects of GSKJ4 in human leukemia U937 cells. We provide evidence that forskolin markedly sensitizes GSKJ4-induced antiproliferative effects through apoptosis induction (caspase-3 activation and PARP cleavage). In addition, we demonstrate that this phenomenon is mediated by cAMP elevation and Protein Kinase A (PKA) involvement, as indicated by the use of PKA inhibitor (KT-5720). In conclusion, our findings provide initial evidence about the efficacy of forskolin/GSKJ4 combination in leukemia cells and suggest a new possible therapeutic approach for AML treatment. Development of resistance to kinase inhibitors remains a clin. challenge. Kinase domain mutations are a common mechanism of resistance in chronic myeloid leukemia (CML), yet the mechanism of resistance in the absence of mutations remains unclear. We tested proteins from the bone marrow microenvironment and found that FGF2 promotes resistance to imatinib in vitro. Fibroblast growth factor 2 (FGF2) was uniquely capable of promoting growth in both short- and long-term assays through the FGF receptor 3/RAS/c-RAF/mitogen-activated protein kinase pathway. Resistance could be overcome with ponatinib, a multikinase inhibitor that targets BCR-ABL and FGF receptor. Clin., we identified CML patients without kinase domain mutations who were resistant to multiple ABL kinase inhibitors and responded to ponatinib treatment. In comparison to CML patients with kinase domain mutations, these patients had increased FGF2 in their bone marrow when analyzed by immunohistochem. Moreover, FGF2 in the marrow decreased concurrently with response to ponatinib, further suggesting that FGF2-mediated resistance is interrupted by FGF receptor inhibition. These results illustrate the clin. importance of ligand-induced resistance to kinase inhibitors and support an approach of developing rational inhibitor combinations to circumvent resistance.
