Antibiotics such as sulfonamides are widely used in agriculture as growth promoters and medicine in treatment of infectious diseases. However, the release of these antibiotics has caused serious environmental problems. In this paper, photocatalytic oxidation technol. was used to degrade sulfadiazine (SDZ), one of the typical sulfonamides antibiotics, in UV illuminated TiO2 suspensions. It was found that TiO2 nanosheets (TiO2-NSs) with exposed (001) facets exhibit much higher photoreactivity towards SDZ degradation compared to TiO2 nanoparticles (TiO2-NPs) with a rate constant increases from 0.017 min-1 to 0.035 min-1, improving by a factor of 2.1. Under the attacking of reactive oxygen species (ROSs) such as superoxide radicals (·Ousc2) and hydroxyl radicals (·OH), SDZ was steady degraded on the surface of TiO2-NSs. Based on the identification of the produced intermediates by LC-MS/MS, possible degradation pathways of SDZ, which include desulfonation, oxidation and cleavage, were put forwards. After UV irradiation for 4 h, nearly 90% of the total organic carbon (TOC) can be removed in suspensions of TiO2-NSs, indicating the mineralization of SDZ. TiO2-NSs also exhibits excellent stability in photocatalytic degradation of SDZ in wide range of pH. Even after recycling used for 7 times, more than 91.3% of the SDZ can be efficiently removed, indicating that they are promising to be practically used in treatment of wastewater containing antibiotics.

Pyruvate dehydrogenase kinases (PDKs) and lactate dehydrogenase A (LDHA) play crucial roles in promotion of glycolysis, thus the inhibition of both enzymes is considered a promising strategy for developing of anticancer therapeutics. Herein, we describe the first discovery of series novel dual inhibitors targeting PDKs and LDHA. We identified 6 hits from a library database containing 485465 compounds through a high-throughput virtual screening assay. Hit-to-lead optimization enabled us to discover two compounds, namely 20e and 20k, which inhibited PDKs with IC50 values of 0.8, and 1.6 μM, resp., and inhibited LDHA with IC50 values of 0.15 and 0.7 μM, resp. Meanwhile, the two compounds reduced A549 cell proliferation with EC50 values of 13.2, and 15.7 μM. Furthermore, 20e and 20k decreased the lactate formation, and increased oxygen consumption, suggesting the two compounds modulated the glucose metabolic pathways in cancer cells.

In contrast to the well-established genomic 5-methylcytosine (5mC), the existence of N6-methyladenine (6 mA) in eukaryotic genomes was discovered only recently. Initial studies found that it was actively regulated in cancer cells, suggesting its involvement in the process of carcinogenesis. However, the contribution of 6 mA in tongue squamous cell carcinoma (TSCC) still remains uncharacterized. In this study, a pan-cancer type anal. was first performed, which revealed enhanced 6 mA metabolism in diverse cancer types. The study was then focused on the regulation of 6 mA metabolism, as well as its effects on TSCC cells. To these aspects, genome 6 mA level was found greatly increased in TSCC tissues and cultured cells. By knocking down 6 mA methylases N6AMT1 and METTL4, the level of genomic 6 mA was decreased in TSCC cells. This led to suppressed colony formation and cell migration. By contrast, knockdown of 6 mA demethylase ALKBH1 resulted in an increased 6 mA level, enhanced colony formation, and cell migration. Further study suggested that regulation of the NF-κB pathway might contribute to the enhanced migration of TSCC cells. Therefore, in the case of TSCC, we have shown that genomic 6 mA modification is involved in the proliferation and migration of cancer cells.

Copper(I)-copper(II) pyrimidinecarboxylate metal-organic framework [(μ-5-PymCO2)4Cu2py2[Cu4I4]] (1; PymCO2H = 5-pyrimidinecarboxylic acid, py = pyridine) was prepared as active and robust catalyst for hydroboration of alkynes, yielding vinylboronates. The hydroboration of alkynes is crucial due to the wide applications in organic synthesis, while such reaction is often completed with low turnover frequency (TOF) value and long reaction time. Therefore, it is very important and necessary that the hydroboration of alkynes is performed with high TOF value, however the corresponding investigations have been never reported hitherto. Herein, a new Cu-organic framework 1 with mixed-valence Cu(I) and Cu(II) blocks was successfully synthesized and employed for the hydroboration of alkynes with bis(pinacolato)diboron (B2Pin2). The MOF 1 displays good thermostability and excellent solvent stability. Catalytic explorations reveal that 1 can serve as a high efficient heterogeneous catalyst for this reaction with a record TOF value of 310 h-1 under mild conditions, and 1 as catalysts which can be recycled at least five times without adding any cocatalysts. Mechanism investigations suggest that the Cu(I) and Cu(II) clusters in the framework of 1 have a synergistic catalytic effect in the hydroboration of alkynes, which can effectively activate the alkyne to react with B2Pin2. The d. functional theory (DFT) calculations explicitly elucidate the reaction pathways, and the results indicate that the Cu(I) and Cu(II) clusters in 1 as the catalytic sites can greatly reduce the Gibbs free energy of the hydroboration of alkyne in different degree, which accounts for the high catalytic activity of 1.
